A Quantitative In Vivo
Measure of CYP3A4 Activity
The Erythromycin Breath Test is a quantitative in vivo
measure of hepatic CYP3A4 activity (cytochrome P450),
the most important drug-metabolizing enzyme present in
the liver. The test provides a phenotypic measurement
of individual variance in hepatic enzymatic activity due
to factors such as disease, diet, concomitant medications
as well as genetic differences.
The Erythromycin Breath Test is simple and quick to perform,
and can be easily added to existing research protocols.
The test is performed by intravenously administering
a trace amount of 14C labeled erythromycin
and then measuring the amount of exhaled 14CO2.
The test does not require special equipment; the
patient simply blows up a single small balloon 20 minutes
after the IV dose injection.
The level of 14CO2 correlates
with the level of hepatic CYP3A4 (cytochrome P450) enzyme
activity.
Metabolic Solutions is the sole manufacturer of Erythromycin
Breath Test Kits and source for laboratory services
to measure breath specimens. Since 1992, Metabolic Solutions
has been the source for kits and analysis in virtually
all published studies involving the Erythromycin Breath
Test.
Metabolic Solutions provides the following full services
that makes it easy to for researchers to employ the
Erythromycin Breath Test in clinical studies:
- Assistance with experimental design / protocols
- Providing standard or custom test kits containing
all the components necessary to administer a erythromycin
breath test. These test kits are ideal for use
at contract research organizations.
- Analysis of the 14CO2 at
our laboratory.
- Custom reporting options.
- Bioanalytical reports are available (including
summary of study data, description of analytical and
Quality Control (QC) methods, QC data, and certification
of results by an independent QA auditor and the Analytical
Project Manger) suitable for FDA submissions.
